Myelin fragment-specific antigens are peptide fragments derived from myelin proteins that act as antigens, triggering immune responses. These fragments are central in the study of autoimmune demyelinating diseases such as multiple sclerosis (MS), where the immune system targets components of myelin in the central nervous system (CNS). Key myelin antigens include Myelin Oligodendrocyte Glycoprotein (MOG) and Proteolipid Protein 1 (PLP1). Specific peptide fragments from these proteins, such as MOG35–55, are commonly used to induce experimental autoimmune encephalomyelitis (EAE), a model for MS. The immunodominant epitopes often reside on exposed loops or domains accessible at the surface of intact myelin sheaths. Autoimmune responses against specific myelin protein fragments lead to inflammation, demyelination, and neurodegeneration.
