Overview

Myelin membrane lipids are the primary structural components of the myelin sheath, a specialized multi-layered membrane that insulates axons in the central and peripheral nervous systems (StatPearls, 2023). Comprising approximately 70-80% of the myelin dry weight, these lipids—including cholesterol, galactocerebroside, and sulfatide—are essential for saltatory conduction, which significantly increases the speed of nerve impulse transmission (PubMed, PMID: 22403187). In neurodegenerative and demyelinating diseases like Multiple Sclerosis, these lipids are often the target of autoimmune attacks or metabolic deficiencies, leading to axonal vulnerability and functional loss (Mayo Clinic, 2024). Therapeutic strategies targeting myelin lipids focus on either protecting existing structures from inflammatory degradation or stimulating oligodendrocyte-mediated remyelination to restore lipid bilayers (Nature Reviews Neurology, 2021). Modern pharmacological approaches include sphingosine-1-phosphate (S1P) receptor modulators like Fingolimod and small molecules like Clemastine that promote the differentiation of oligodendrocyte precursor cells into mature, lipid-producing cells (The Lancet, 2017).
