Myelin protein zero-like protein 3 (MPZL3) is a single-pass transmembrane glycoprotein characterized by an immunoglobulin (Ig)-like variable (V)-type domain, predicted to function as a cell adhesion molecule[1][3]. It is encoded by the MPZL3 gene at chromosome 11q23.3 and is implicated in various cellular processes including cell–cell adhesion, cell differentiation, and antigen binding[1][3]. MPZL3 plays roles in energy and lipid metabolism, circadian rhythm regulation, and immune system function[1]. It is frequently overexpressed in cancers such as breast carcinoma, correlates with poor prognosis, and is associated with tumor immune cell infiltration and drug sensitivity/resistance[1]. MPZL3 expression level may serve as both a prognostic and predictive biomarker in oncology. While several drugs modulate response in tumors with altered MPZL3 expression, no clinically approved agents directly target MPZL3 to date[1]. Disease associations include cancer, seborrheic dermatitis, and metabolic disorders[5].
