Overview

Myeloid-associated differentiation marker (MYADM) is a multi-pass membrane protein belonging to the MAL protein family and uniquely characterized by containing two MARVEL domains and spanning the membrane eight times. MYADM is widely expressed in human tissues, localizing mainly to the plasma membrane, where it plays a critical role in organizing lipid raft microdomains. These specialized domains modulate membrane condensation and protein sorting, which are essential for processes including cell migration (by enabling proper Rac1 GTPase localization), cytoskeletal regulation, and immune cell signaling. MYADM is necessary for surfactant protein A (SP-A)–mediated induction of eosinophil apoptosis, suggesting a significant role in airway homeostasis and allergic inflammation such as asthma. Changes in MYADM expression have additional links to inflammatory diseases, metastatic cancer, and hypertension. While not a classic druggable receptor, MYADM functions as a critical organizer of membrane platforms and mediator of cell signaling, potentially making it a novel therapeutic or biomarker target in immune and inflammatory conditions
