Myeloid differentiation primary response protein 88 (MyD88) is a cytosolic adaptor protein that plays a central role in both innate and adaptive immune responses, primarily by mediating signal transduction from Toll-like receptors (TLRs) and interleukin-1 receptor (IL-1R) family members. Upon activation of TLRs or IL-1R/IL-18R, the TIR domain of MyD88 interacts with the corresponding receptor’s TIR domain, recruiting IL‑1 receptor-associated kinases (IRAKs) and activating downstream signaling cascades, including NF‑κB, MAPKs, and AP‑1. Mutations in MYD88, particularly L265P, are implicated in lymphomas. It is a target for drug discovery efforts focused on inhibitors/modulators for various diseases involving dysregulated innate immunity.
