Overview

Myocardin-related transcription factor A (MRTF-A) is a transcriptional coactivator that functions primarily by interacting with the serum response factor (SRF) to regulate gene expression involved in cytoskeletal dynamics, myogenic differentiation, and cellular response to growth factors. MRTF-A serves as a key bridge between Rho-actin signaling and gene transcription by sensing changes in monomeric and filamentous actin; under conditions of actin polymerization, MRTF-A translocates to the nucleus, where it cooperates with SRF to activate a subset of genes critical for cytoskeletal and muscle cell functions. It is expressed widely in embryonic and adult tissues and is involved in stem cell fate decisions such as the balance between adipogenic and osteogenic differentiation. Alterations in MRTF-A function or expression, particularly due to chromosomal translocation, have been implicated as oncogenic events in some forms of leukemia, and its dysregulation is linked to cardiovascular disorders. MRTF-A is considered a plausible therapeutic target in diseases related to aberrant actin dynamics, smooth muscle proliferation, and stem cell differentiation, and it can be pharmacologically modulated by small molecule inhibitors such as CCG-1423 and CCG-100602[1][2][3][4][5].
