Overview

MyoD family inhibitor domain-containing protein (MDFIC) is a multifunctional modulatory protein defined by a cysteine-rich C-terminal I-mfa domain[1][3][5]. It regulates transcription by interacting with and sequestering transcription factors (notably the Zic family) in the cytoplasm, as well as modulating the activity of cellular and viral promoters through direct interaction with viral proteins (e.g., HIV Tat, HTLV-I Tax) and signaling pathways such as Wnt[4][5][7]. MDFIC influences epigenetic regulation by interacting with histone demethylase JMJD1A, thereby affecting expression of tumor suppressor genes and cancer cell proliferation. Non-transcriptional roles include regulation of lymphatic vessel formation (with biallelic loss-of-function mutations causing central conducting lymphatic anomaly) and modulation of Piezo mechanosensitive channels as an auxiliary subunit. Alternative translation and splicing generate distinct isoforms with differential subcellular localization and activity[1][3][5][7]. There is currently no direct evidence that MDFIC functions as a classical receptor, transporter, or enzyme drug target, nor are there any known drugs or biomarker uses in clinical practice.
