Myogenic factor 6 (MYF6)

Target
MYF6
Molecular classification
Transcription factor, basic helix-loop-helix family

Overview

Myogenic factor 6 (MYF6) is a basic helix-loop-helix (bHLH) transcription factor specifically expressed in skeletal muscle, with a pivotal role in the regulation of muscle differentiation (myogenesis) and muscle regeneration[1][2][3]. It is part of the myogenic regulatory factor (MRF) family, which also includes MYOD1, MYOG, and MYF5. MYF6 is involved in transcriptional activation of a range of muscle-specific genes and has a unique function in regulating myokine expression in muscle fibers, impacting the maintenance of the adult muscle stem cell (satellite cell) pool[2]. Mutations in MYF6 are associated with autosomal dominant centronuclear myopathy and have been linked to various inherited muscle disorders, but no approved drugs currently target MYF6 directly[1][3].

Safety considerations

Null mutations result in loss of regulation of muscle stem cells, leading to premature differentiation and exhaustion of the stem cell poolMutations can cause congenital myopathies with varying severity
