Myomesin-3 (MYOM3) is a member of the myomesin protein family, predominantly found in the M-band region of the sarcomere in striated muscle cells. It is primarily expressed in skeletal muscle fibers of intermediate contraction speed and in specific regions of the adult heart, including the left ventricle and atrial appendage. Structurally, MYOM3 consists of a unique N-terminal domain, followed by two immunoglobulin-like domains, five fibronectin type III domains, and five additional immunoglobulin-like domains. It provides mechanical stability by cross-linking myosin and titin filaments, contributing to sarcomere elasticity and muscle contraction mechanics. MYOM3 forms homodimers but does not heterodimerize with other myomesin family members. Although its tissue-specific expression suggests important mechanical and possibly signaling roles within certain muscle types, MYOM3 remains poorly characterized in terms of molecular interactions and disease relevance. Its function and disease associations are still under investigation, though it is considered part of a protein family increasingly linked to muscle and cardiovascular disease pathology.
