Overview

Myosin-1E (MYO1E) is a member of the nonmuscle class I myosin family of unconventional myosins, functioning as an actin-dependent molecular motor protein[3][4][6]. It has a characteristic long tail domain involved in crosslinking actin filaments and localizes to the cytoplasm, where it mediates intracellular movement and membrane trafficking, particularly in clathrin-mediated endocytosis and actin cytoskeleton dynamics[1][3][4][7]. MYO1E directly interacts with proteins such as focal adhesion kinase (FAK) to regulate cell adhesion, gene expression, and cancer-related processes[2], and plays a regulatory role in podocyte structure and function, with mutations causing genetic kidney disease such as focal segmental glomerulosclerosis[3][5]. It also participates in actin assembly, vesicle trafficking, and influences cell motility and rigidity, with broader implications in cancer metastasis, fibrosis, and tissue repair[2][3].
