Overview

Myosin-binding protein H (MYBPH) is a muscle protein that binds to myosin, localizes primarily to the thick myofilaments of the sarcomere’s A-band and M band, and is implicated in the structural organization of muscle tissue[1][3][5][6]. MYBPH is closely related to cardiac myosin-binding protein C and can compensate for its function in cardiac contractility[2]. In non-muscle contexts, MYBPH regulates actomyosin dynamics by directly inhibiting ROCK1-mediated phosphorylation and the assembly of non-muscle myosin heavy chain IIA, which impacts cell migration and metastasis[1]. Increased MYBPH expression in skeletal muscle is linked to early disturbances in actomyosin dynamics observed in amyotrophic lateral sclerosis[1]. Genetic variants in MYBPH modulate the severity of cardiac hypertrophy in hypertrophic cardiomyopathy patients[1][2]. At present, there are no known drugs specifically targeting MYBPH, nor is it considered a classic therapeutic target such as a receptor, enzyme, or transporter, but it has emerging interest as a disease modifier and a potential biomarker in certain contexts[1][2][3].
