Myosin heavy chain 14 (MYH14) encodes a conventional non-muscle myosin protein, specifically the heavy chain of non-muscle myosin IIC.[2][3] As an actin-dependent motor protein, MYH14 functions as a molecular motor that hydrolyzes ATP and generates physical force to regulate diverse cellular processes including cytokinesis, cell motility, cell polarity, and specialized functions such as secretion and receptor capping.[2][5] The protein assembles into bipolar filaments and interacts with actin to produce contractile force essential for cellular architecture and function.[3] MYH14 is particularly critical in auditory cells, where it maintains inner ear cell structure and hearing function.[3] Mutations in the MYH14 gene result in autosomal dominant hearing impairment and have been associated with other clinical phenotypes including peripheral neuropathy, myopathy, and hoarseness.[2] Unlike some myosin family members, MYH14 is not currently recognized as an established therapeutic target for drug development, and no pharmacological agents are known to specifically modulate its activity.
