Myosin heavy chain 7B (MYH7B) is an evolutionarily ancient member of the sarcomeric myosin family, encoding a slow-twitch myosin II heavy chain that participates in the architecture and function of striated muscle, especially in specialized muscle tissues and, to a limited extent, in certain nonmuscle environments. The protein consists of a globular motor domain responsible for ATP hydrolysis and actin binding, and a rod-like tail that promotes dimerization. In mammals, alternative splicing largely prevents MYH7B protein expression in adult cardiac and skeletal muscles, limiting its presence to specialized muscle groups, though it is abundant in cardiac and skeletal muscles of some non-mammalian species such as reptiles. MYH7B is involved in slow and energy-conserving motor activity, potentially contributing to evolutionary muscle specialization. Mutations in the family of myosin heavy chains (though more commonly in MYH7 than MYH7B) are associated with several myopathies and cardiomyopathies in humans. There are currently no known drugs directly targeting MYH7B nor is it recognized as a conventional therapeutic target.
