Overview

Myosin heavy chain gene cluster antisense RNA (MYHAS), also known as LINC-MYH and linc-MYH, is a long non-coding RNA (lncRNA) located on human chromosome 17p13.1[3][10]. It is transcribed antisense to the myosin heavy chain gene cluster and is classified as a long intergenic non-coding RNA (lincRNA)[10][11]. MYHAS plays a regulatory role in muscle gene expression by suppressing slow-type myosin heavy chain (Myh) gene expression and stimulating fast-type Myh gene expression (such as Myh1, Myh2, and Myh4) through enhancer-promoter looping mechanisms[9]. This RNA is also predicted to possess primary miRNA binding activity, suggesting possible involvement in microRNA-mediated regulation[5][11]. Variants or dysregulation of MYHAS have been associated with cancer outcomes, notably serving as a protective prognostic marker in laryngeal cancer[9]. There is no evidence at present that MYHAS is directly targeted by therapeutic drugs or acts as a classic therapeutic target such as a receptor, enzyme, or transporter[7].
