Myosin light-chain kinase 2 (MYLK2), also known as skeletal muscle myosin light chain kinase (skMLCK or MLCK2), is a calcium/calmodulin-dependent serine/threonine protein kinase. It phosphorylates the regulatory light chain of myosin II, initiating muscle contraction in skeletal muscle fibers. MYLK2 plays a critical role in regulating actin-myosin crossbridge cycling. Mutations in MYLK2 are linked to midventricular hypertrophic cardiomyopathy.
