Overview

Myosin light chain kinase 3 (MYLK3) is a cardiac-specific serine/threonine protein kinase that phosphorylates myosin regulatory light chain 2 (MLC2) in a calcium/calmodulin-dependent manner, potentiating force generation and contractility in cardiac myocytes[1][2][4][5]. Expression of MYLK3 is highly specific to the heart, and it plays a central role in sarcomere organization and normal cardiac muscle function[5]. Mutations in the MYLK3 gene disrupt MLC2 phosphorylation and have been linked to familial dilated cardiomyopathy, emphasizing its critical role in cardiac health[3][5]. There are currently no approved drugs directly targeting MYLK3, and its modulation is associated with potential cardiac safety concerns due to its essential function in myocardial contractility and structure[3][5].
