Myosin regulatory light chain 12B (MYL12B) is a protein encoded by the MYL12B gene in humans. It is a regulatory subunit of nonmuscle and smooth muscle myosin II, functioning primarily to regulate contractile activity in muscle and nonmuscle cells through phosphorylation by myosin light chain kinase. Upon phosphorylation, MYL12B increases actin-activated myosin ATPase activity, facilitating myosin II filament assembly and cytoskeletal rearrangements. Its main biological roles include regulation of muscle contraction, actin cytoskeleton organization, cell division (cytokinesis), cell migration, and receptor capping. MYL12B is implicated in certain pathophysiological states such as dilated cardiomyopathy when its regulation or function is disrupted. There are currently no drugs known to directly target MYL12B, and it is not widely regarded as a direct therapeutic target, but it plays significant roles in signal transduction underlying cellular motility and division[1][2][3][7].
