MYO5B
Molecular classification
Actin-based molecular motor protein, Motor protein, Cytoskeletal protein, Other
01

Overview

Myosin-Vb is an actin-based molecular motor protein encoded by the MYO5B gene, widely expressed in epithelial and other tissues. It is critical for trafficking and recycling of membrane proteins (especially apical and basolateral in polarized cells), maintenance of cell polarity, and intracellular vesicle transport. Loss of MYO5B function disrupts the formation and maintenance of microvilli on enterocytes, causing microvillus inclusion disease, a life-threatening condition characterized by severe neonatal diarrhea and malabsorption. It also plays roles in hepatocyte protein trafficking and has been connected to forms of genetic cholestasis and potentially memory through synaptic regulation[1][2][3][4][5][6][7].

Safety considerations

Genetic loss-of-function leads to severe diarrhea, malnutrition, dehydration, liver dysfunction, and possibly developmental disorders; no notable pharmacological safety issues since not a direct drug target[1][3][7].
