Overview

Myotrophin is a small (12–13 kDa), ankyrin repeat-containing protein encoded by the MTPN gene that plays a significant role in the initiation and progression of cardiac muscle hypertrophy[1][2][3][4][5]. It promotes the growth of cardiomyocytes (not proliferation), likely through interaction with the F-actin capping protein complex, inhibiting its function, and allowing actin filament extension[2][3][4]. Myotrophin also regulates the NF-kappa-B pathway by promoting dimerization of its subunits, impacting a range of cellular stress and survival pathways[3][4]. Alternative translation from the same gene produces leucine zipper protein 6, linked to myeloproliferative disease, but this is distinct from the canonical myotrophin function[3]. Myotrophin has been most intensively studied as a candidate cardiac hypertrophy protein and is being explored for its roles as a disease biomarker and a potential pharmacological target in heart failure and related conditions[1][3][5][7].
