Overview

Myotubularin-related protein 11 (MTMR11) is a member of the myotubularin family of lipid phosphatases but is classified as a *pseudophosphatase* due to absence of a conserved active site cysteine residue required for enzymatic activity[3]. The MTMR protein family modulates cellular levels of phosphoinositides involved in membrane trafficking, cell proliferation, and autophagy[1]. MTMR11 is believed to function largely through protein–protein interactions with other MTMR family proteins rather than direct phosphatase activity. MTMR11 is reported as upregulated by HER2 and may be associated with malignant tumor cell proliferation. There are limited and indirect reports linking it to *cisplatin resistance*, though strong functional evidence is lacking. Overall, MTMR11 is an adaptor or regulatory protein with likely roles in cellular signaling, but it is not an established therapeutic target or direct drug-interaction partner[3][1][5].

Mechanism of action

Not applicable; no known drugs directly target MTMR11. Family members may interact with the PI3K/AKT/mTOR axis that is relevant for cisplatin resistance and chemotherapeutic response. Cisplatin resistance mechanisms are diverse and include drug efflux, DNA repair, apoptotic pathways, and the PI3K/AKT/mTOR signaling axis, but direct involvement of MTMR11 is not established.
