Myotubularin-related protein 3 (MTMR3) is a member of the myotubularin dual specificity protein phosphatase family, encoded by the MTMR3 gene in humans[1][6][9]. It acts primarily as a lipid phosphatase, specifically hydrolyzing phosphatidylinositol 3-phosphate (PI(3)P) and PI(3,5)P2 lipids, thereby playing a critical role in membrane trafficking, autophagy, vesicle transport, and signal transduction[1][2][9][10]. MTMR3 has a multidomain structure, including a FYVE domain for lipid binding and a PH-GRAM domain, and is involved in diverse cellular processes such as cell proliferation, differentiation, migration, and cytoskeleton dynamics[1][2][10]. Dysregulation of MTMR3 has been implicated in cardiovascular diseases, preeclampsia, and possibly other metabolic and proliferative disorders[2]. There are currently no approved drugs directly targeting MTMR3.
