Overview

Myotubularin related protein 6 (MTMR6) is a lipid phosphatase enzyme that targets phosphoinositides, specifically dephosphorylating phosphatidylinositol 3-phosphate and phosphatidylinositol 3,5-bisphosphate. It plays a key regulatory role in vesicular trafficking (including the Golgi-ER and endocytic pathways), autophagy, and protein transport. MTMR6 forms a heterodimeric complex with MTMR9, which is essential for its function and subcellular localization. It is found in the cytoplasm and perinuclear regions, where its localization is partially regulated by the GTPase Rab1B. MTMR6 negatively regulates certain transport processes, such as ER-Golgi transport, and is required for late macropinocytosis steps. Loss or dysfunction of MTMR6, as observed in some genetic diseases, disrupts intracellular trafficking and may contribute to myopathic or hematological disorders.
