Overview

Myotubularin related protein 8 (MTMR8) is a member of the myotubularin-related family of lipid phosphatases, specifically classified in the MTMR6 subgroup[1][3]. MTMR8 functions as a dual-specificity phosphatase with the ability to dephosphorylate phosphatidylinositol 3-phosphate and phosphatidylinositol 3,5-bisphosphate, regulating the cellular pools of these key lipid signaling molecules[1][2][3]. Through these activities, MTMR8 modulates autophagy (often in complex with the catalytically inactive MTMR9), vesicle trafficking, and cell proliferation[2][3]. It has also been implicated in disease states such as autism spectrum disorder, rare skeletal dysplasia (osteopathia striata with cranial sclerosis), and potentially contributes to cardiovascular and oncologic conditions[1][3]. MTMR8 shares structural domains typical of this protein family, including a phosphoinositide-binding domain and a SET-interacting domain, supporting its roles in both lipid signaling and protein-protein interactions[1][3]. To date, there are no commonly referenced drugs or clinical biomarkers targeting MTMR8 directly.
