Myotubularin-related protein 9 (MTMR9) is a member of the myotubularin family that, unlike most myotubularin proteins, lacks the conserved dual-specificity phosphatase domain required for phosphatase activity[1][3]. Instead, MTMR9 acts as an adapter protein, forming complexes with catalytically active myotubularin phosphatases such as MTMR6, MTMR7, and MTMR8. These complexes dramatically enhance the enzymatic activity and stability of partner phosphatases, alter their substrate specificity, and regulate cellular lipid signaling, thereby influencing cell proliferation, apoptosis, and autophagy[2][3]. MTMR9 does not directly catalyze the dephosphorylation of phosphatidylinositol phosphates but is essential in modulating phosphoinositide metabolism through its interactions. Genetic studies have linked MTMR9 to obesity and several rare inherited diseases, but no direct therapeutic strategies or drug interactions targeting MTMR9 are currently reported[2][3].
