Myozenin-2 (MYOZ2) is a muscle-specific sarcomeric protein localized at the Z-disc of cardiac and slow-twitch skeletal muscle fibers. It serves as an anchor for calcineurin, a calcium/calmodulin-dependent phosphatase, tethering it to alpha-actinin and other structural components at the sarcomere. By inhibiting calcineurin signaling, MYOZ2 suppresses pathological cardiac hypertrophy and regulates muscle fiber type specification and development. Mutations in MYOZ2 cause familial hypertrophic cardiomyopathy type 16 and have been implicated in broader muscle and heart pathologies. No drugs are known to directly target MYOZ2, but its function affects the progression and drug response in heart disease. MYOZ2 is not classified as a receptor, enzyme, transporter, or ion channel, but rather as a structural and signaling modulator in muscle tissue.
