Overview

Myozenin-3 (MYOZ3) is a muscle-specific, Z-disc–associated intracellular protein expressed predominantly in fast-twitch skeletal muscle, and to a lesser extent in cardiac muscle[1][2][3]. It functions primarily as a structural and signaling adapter, interacting with calcineurin, α-actinin, γ-filamin, telethonin, and LDB3/ZASP at the sarcomere[2][3]. MYOZ3 modulates calcineurin signaling, which is important for determining muscle fiber type and muscle cell growth[1][3]. Unlike typical molecular therapeutic targets like receptors or enzymes, MYOZ3 is not currently considered a classic therapeutic target for drugs, though it has been investigated as a candidate gene in neuromuscular and congenital contracture syndromes[2][3]. There are no established small-molecule or biological drugs that target MYOZ3, and no known safety or biomarker data regarding its modulation in disease.
