The M013 protein is a viral immunomodulatory factor encoded by the Myxoma virus, a member of the Poxviridae family. It functions as a viral pyrin-only protein (vPOP) that effectively subverts the host's innate immune response by targeting two critical signaling pathways: the inflammasome and the NF-kappaB pathway. Specifically, M013 binds directly to the host protein ASC-1 (apoptosis-associated speck-like protein containing a CARD), preventing the assembly of the inflammasome complex and the subsequent activation of caspase-1 and release of pro-inflammatory cytokines like IL-1beta and IL-18. Additionally, it interacts with the NF-kappaB1 (p105) subunit to inhibit NF-kappaB-mediated transcription of inflammatory genes. Due to its potent anti-inflammatory properties, M013 is being investigated as a therapeutic agent, often delivered via viral vectors like AAV, for the treatment of inflammatory and autoimmune conditions such as uveitis and potentially as part of oncolytic virotherapy strategies.
