N-acetyl-D-glucosamine (GlcNAc) is a naturally occurring amino sugar and a derivative of glucose that serves as a fundamental building block in various biological structures (Biology Online, 2021). It is a primary component of bacterial peptidoglycan, fungal chitin, and mammalian glycosaminoglycans such as hyaluronic acid and keratan sulfate (Wikipedia, 2024). Beyond its structural roles, GlcNAc is a key player in cell signaling through O-GlcNAcylation, a dynamic post-translational modification of nuclear and cytoplasmic proteins that regulates their function in response to nutrient availability (NIH, 2015). In a therapeutic context, GlcNAc is investigated for its anti-inflammatory and chondroprotective properties, particularly in the treatment of osteoarthritis and inflammatory bowel disease, where it supports tissue repair and mucosal barrier integrity (Patsnap, 2024). Recent research also highlights its potential in autoimmune diseases like multiple sclerosis by modulating N-glycan branching and T-cell activity (NIH, 2023). It is also used in cosmetic formulations to improve skin hydration and reduce hyperpigmentation (NIH, 2024).
N-acetyl-D-glucosamine acts as a substrate for the biosynthesis of glycosaminoglycans and proteoglycans, supporting tissue repair and mucosal integrity (Patsnap, 2024). It also modulates immune responses by inhibiting pro-inflammatory cytokines like IL-6 and TNF-alpha and reducing nitric oxide production (Darwin Nutrition, 2025). In autoimmune contexts, it enhances N-glycan branching on T-cell receptors, which suppresses overactive immune signaling (NIH, 2023). Additionally, it serves as the donor for O-GlcNAcylation, a post-translational modification that regulates protein function and gene expression (Wikipedia, 2024).
