Overview

The Acetate / N-acetylaspartate (NAA) system is a specialized metabolic pathway in the central nervous system that facilitates the transfer of acetate from neurons to oligodendrocytes for myelin synthesis. NAA is synthesized in neuronal mitochondria by the enzyme N-acetyltransferase 8-like (NAT8L) from acetyl-CoA and aspartate. It is then transported to oligodendrocytes, where it is hydrolyzed by the enzyme aspartoacylase (ASPA) into acetate and aspartate. This system is critical for brain development and maintenance; its disruption is the hallmark of Canavan disease, where ASPA deficiency leads to toxic NAA accumulation and spongy brain degeneration. Beyond leukodystrophies, the system is implicated in cancer, where NAT8L overexpression supports tumor growth by providing acetyl-CoA for lipid synthesis and histone acetylation. Therapeutic strategies include gene therapy to restore ASPA function, acetate supplementation to provide lipid precursors, and NAT8L inhibitors to reduce NAA levels in disease states.
