Overview

N-acylsphingosine amidohydrolase (non-lysosomal ceramidase) 2B (ASAH2B) is a putative member of the ceramidase enzyme family, predicted to catalyze the hydrolysis of ceramides into sphingosine and fatty acids as part of cellular lipid metabolism, although direct enzymatic activity for human ASAH2B remains unconfirmed[1][5][6]. ASAH2B is believed to reside in membrane structures within the cell, including likely associations with endosomal and vesicular membranes[2][3][6]. The ASAH2B gene locus also gives rise to a long noncoding RNA (lnc-ASAH2B-2) which modulates cellular signaling pathways, notably activating mTOR in breast cancer cells, thereby promoting cell proliferation and resistance to the drug everolimus[1]. Altered expression of ASAH2B/related family members has been observed in neurodegenerative conditions such as Alzheimer’s disease and with aging, suggesting a context-dependent regulatory role in both cancer and neurobiology[1][3][5]. However, specific therapeutic targeting and validated molecular drug interactions for ASAH2B are not currently established, distinguishing it from the related ASAH2 enzyme, which is more clearly implicated in pathological conditions[3][5][6].
