Overview

N-glycolylneuraminic acid-containing ganglioside GM3 (NGcGM3) is a sialic acid-containing glycosphingolipid found in cell membranes. It is characterized by the presence of N-glycolylneuraminic acid (Neu5Gc) as its sialic acid moiety. Humans cannot synthesize Neu5Gc, but it can be incorporated from dietary sources. NGcGM3 is selectively expressed in many human cancers, making it a promising target for immunotherapies.

Mechanism of action

Monoclonal antibodies (e.g., 14F7) target NGcGM3 for antibody-dependent cell-mediated cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). CAR-T therapies can be engineered to recognize and eliminate NGcGM3-expressing cells.
