Overview

The N-methyl-D-aspartate (NMDA) receptor is a subtype of ionotropic glutamate receptors, which are the primary mediators of excitatory neurotransmission in the central nervous system. It plays a critical role in synaptic plasticity, learning, memory formation, and overall brain development. Abnormal NMDA receptor function has been implicated in various neurological disorders. Hypofunction is associated with schizophrenia spectrum disorders due to impaired synaptic transmission/plasticity, while hyperfunction/excessive activation can lead to excitotoxic neuronal death seen in stroke/traumatic brain injury.
