Overview

The N-methyl-D-aspartate receptor 3A subunit (GluN3A) is a component of NMDA receptors, which are ionotropic glutamate receptors mediating excitatory neurotransmission. Unlike canonical NMDA receptors, GluN1-GluN3 receptors are activated solely by glycine. They exhibit reduced single-channel conductance, low calcium permeability, and low voltage-dependent sensitivity to magnesium block. GluN3A plays specialized roles in neuronal signaling distinct from typical synaptic plasticity functions, including regulation of neuronal excitability, modulation of circuit function, involvement in dendritic spine development, and participation in PPP2CB-NMDAR mediated signaling pathways.
