Overview

The N-methyl D-aspartate receptor subtype 2B (NR2B or GluN2B) is a protein subunit that forms part of the NMDA receptor complex, a type of ionotropic glutamate receptor. NMDA receptors are heterotetrameric complexes typically composed of two obligatory NR1 (GluN1) subunits and two regulatory NR2 subunits (NR2A, NR2B, NR2C, or NR2D). The GluN2B/NR2B subunit contributes to the agonist binding site for glutamate and modulates channel properties. It plays a role in synaptic plasticity, learning, memory formation, central sensitization in chronic pain, and modulation of feeding behaviors. Mutations or dysregulation in GRIN2B have been implicated in various neurological disorders including intellectual disability, epilepsy syndromes, neurodevelopmental disorders such as autism spectrum disorder (ASD), schizophrenia risk modulation, chronic pain syndromes due to central sensitization mechanisms mediated by increased activity/expression at synapses involved in pain processing circuits.
