The N-myc downstream regulated gene 1 protein (NDRG1) is a cytoplasmic and, under stress, nuclear protein involved in cellular stress response, cell proliferation, differentiation, vesicular trafficking, and apoptosis. NDRG1 acts as a tumor suppressor in multiple cancers by inhibiting proliferation and metastasis, and is crucial for Schwann cell myelination. It is implicated in hypoxia adaptation and is upregulated by stress, particularly hypoxia and certain metals. Mutations in NDRG1 are causative for Charcot-Marie-Tooth disease type 4D, a demyelinating neuropathy. NDRG1 influences pathways including p53-mediated apoptosis, PI3K, c-Src, and WNT signaling, and stabilizes DNA repair proteins such as MGMT. Its expression and subcellular localization are highly responsive to environmental and metabolic stressors
Not established for direct-acting drugs. Some mechanisms for experimental modulation include inhibition of hypoxia-regulated pathways or induction via heavy metal exposure (e.g., nickel, cobalt) or regulation through tumor suppressor signaling (e.g., p53)
