N-terminal EF-hand calcium-binding protein 1 (NECAB1) is a member of a small family of neuronal proteins characterized by an N-terminal EF-hand calcium-binding domain and a C-terminal antibiotic biosynthesis monooxygenase (ABM) domain[1][2][3][5]. NECAB1 is primarily expressed in the central nervous system, including the cortex, hippocampus, and spinal cord, where it is localized to both cytoplasm and nucleoplasm of select neuronal subpopulations, especially interneurons[1][3][5]. It was originally identified as a binding partner of synaptotagmin-1, implicating it in the regulation of calcium-dependent protein-protein interactions important for neurotransmitter vesicle release and neuronal signaling[1][3]. Though it is a calcium sensor protein, its precise physiological and pathophysiological roles are still being elucidated. Current data suggest NECAB1 does not function as a classical therapeutic "target" such as a druggable receptor, enzyme, or channel, and no drugs are known to modulate its function clinically. There is ongoing research into its potential involvement in neurological disorders, but disease association remains preliminary[1].
