N-terminal EF-hand calcium-binding protein 2 (NECAB2) is a neuronal cytoplasmic and membrane-associated protein, prominently expressed in select brain regions such as the telencephalon, habenula, and cerebellum[1]. NECAB2 is characterized by a single N-terminal EF-hand calcium-binding domain, a centrally located NECAB homology region (NHR), and a C-terminal ABM motif or DUF176 domain[1][2]. This protein is evolutionarily conserved among vertebrates and is localized peri-synaptically in subsets of both glutamatergic and GABAergic neurons[1]. NECAB2 modulates synaptic signaling and has a proven role in regulating the signaling of G protein-coupled receptors, including interaction with metabotropic glutamate receptor 1 (mGluR1), and helps to regulate psychomotor and social behavior[1]. Genetic studies link NECAB2 to autism risk, and proteomic evidence shows its interaction with several autism risk genes[1]. While NECAB2 is vital for neuronal signaling and behavior, it is not classified as a therapeutic target, receptor, enzyme, transporter, or typical druggable class at present[1][2].
