Protein N-terminal Methyltransferase 1 (NTMT1) is a protein methyltransferase that catalyzes the transfer of a methyl group from S-adenosyl-L-methionine (SAM) to the α-amino group at the N-terminus of specific proteins with an exposed [Ala/Gly/Ser]-Pro-Lys motif. It plays a critical role in regulating protein-DNA interactions, mitosis, chromosome segregation, and DNA repair processes. NTMT1 is overexpressed in various cancers and is considered a potential anticancer target. Inhibition of NTMT1 leads to mitotic defects and impaired DNA repair.
