N6-methyladenosine-containing RNA (m6A-containing RNA)

Target
m6A-containing RNA
Molecular classification
Other (epitranscriptomic RNA modification)
01

Overview

N6-methyladenosine-containing RNA refers to RNA molecules in which certain adenosine nucleotides are chemically modified by methylation at the nitrogen-6 position (m6A). This epitranscriptomic mark is reversible and regulates almost every stage of RNA metabolism, including splicing, export, translation, and decay, by influencing the binding of specific proteins ("readers," "writers," and "erasers") that control mRNA and non-coding RNA fate. While the m6A modification is not itself a drug target, its regulatory proteins are emerging therapeutic targets, particularly in cancer and other diseases involving RNA dysregulation[1][2][3][4][7][8].

Mechanism of action

Not applicable directly. For drugs targeting the pathway: inhibition of m6A methyltransferase (writer) activity or demethylase (eraser) activity, altering m6A abundance and downstream gene expression[3][7].

Safety considerations

Not directly applicable, but targeting the m6A pathway may risk broad off-target effects due to central roles in RNA metabolism and cell function[3][8]

Interacting drugs

None directly

Biomarkers

High or low m6A modification levels in specific RNAs can serve as biomarkers in certain cancers and other diseases, but there is no single standardized m6A RNA biomarker[6][7]
