Overview

The N7 position of guanine is the most nucleophilic site within the DNA double helix, located in the major groove where it is highly accessible to electrophilic molecules (PubChem, 2024). It serves as the primary target for several major classes of chemotherapy, including platinum-based agents and nitrogen mustard alkylating agents (StatPearls, 2023). When these drugs bind covalently to the N7 position, they create bulky DNA adducts or cross-links that physically obstruct the machinery required for DNA replication and gene expression (PubMed, 2021). This interference is particularly lethal to rapidly dividing malignant cells, which often have compromised DNA repair mechanisms or high levels of replicative stress (NIH, 2022). However, because these agents also damage DNA in healthy tissues, they are associated with significant clinical toxicities such as bone marrow suppression and the long-term risk of developing secondary cancers (Wikipedia, 2023).

Mechanism of action

Drugs targeting the N7 position of guanine act as electrophiles that form covalent adducts with the nucleophilic nitrogen atom. This process, known as alkylation or platination, results in DNA cross-linking (both intrastrand and interstrand) that physically blocks DNA polymerase and RNA polymerase. The resulting replication stress and transcriptional inhibition activate the DNA damage response, ultimately leading to apoptosis in susceptible cells (StatPearls, 2023; NIH, 2022).
