Overview

Naïve T cells are mature T lymphocytes that have differentiated in the thymus and have not yet encountered their specific antigen in peripheral tissues. Upon antigen recognition by antigen-presenting cells (APCs), naïve T cells are activated through their T cell receptors (TCR), co-stimulatory molecules (such as CD28), and cytokine signals, leading to their differentiation into effector and memory T cells. This activation is essential for initiating adaptive immune responses against previously unencountered pathogens. Naïve T cells are characterized by specific surface markers (CD45RA, CD62L, CCR7, CD127, CD132), and their numbers and functionality decline with age (immunosenescence), resulting in diminished ability to mount new immune responses. Although not a direct molecular target, naïve T cell populations are manipulated in immunotherapy strategies such as adoptive cell therapy or cancer immunotherapy to enhance immune recognition of tumors. Failure to maintain naïve T cell quiescence or homeostasis may contribute to autoimmune diseases, reduced immunity, and cancer.
