The Na-K-Cl cotransporter (NKCC) is a membrane transport protein responsible for the secondary active transport of sodium (Na⁺), potassium (K⁺), and chloride (Cl⁻) ions into cells. There are two main isoforms: NKCC1, ubiquitously expressed and involved in fluid secretion, and NKCC2, primarily in the kidney's thick ascending limb, responsible for renal salt reabsorption. Both are inhibited by loop diuretics and play critical roles in maintaining cell volume and regulating salt/fluid balance.
Loop diuretics like bumetanide and furosemide bind directly to the NKCC transporter protein, inhibiting the co-transport of sodium, potassium, and chloride ions.
