Peptide NA17-A2 antigen (NA17-A2)

Target
NA17-A2
Molecular classification
Other (Tumor-associated antigen), Peptide antigen presented by HLA-A2
01

Overview

The NA17-A2 antigen is a nonamer peptide (sequence: VLPDVFIRC) encoded by an intron of the N-acetylglucosaminyltransferase V (MGAT5) gene, not typically expressed in normal tissues but found in a subset of melanoma and other tumor cells. It is processed and presented on the surface of HLA-A2-positive tumor cells, making it a target for immunotherapeutic approaches in cancer. The antigen has been used in clinical trials as a component of peptide vaccines and for monitoring tumor antigen-specific CD8+ T cell responses in patients with melanoma. Induction of NA17-A2-specific immune responses has been demonstrated in vaccine studies, and its specificity helps minimize autoimmune risk.

Mechanism of action

Induction of specific cytotoxic T lymphocytes (CD8+ T cells) recognizing NA17-A2 peptide presented on HLA-A2-positive tumor cells, leading to immune-mediated tumor cell killing

03

Biological functions

Immune response stimulation (as a T cell epitope)
04

Disease associations

Cancer (primarily melanoma)
05

Safety considerations

Common safety concerns for peptide cancer vaccines include limited immunogenicity, risk of autoimmunity (theoretical), and immunotherapy-related adverse events (as seen in other T cell–based therapies); specific major safety liabilities for NA17-A2 are not widely reported in the literature

07

Biomarkers

Detection of NA17-A2-specific T cell responses (e.g., tetramer staining to monitor immune activation in vaccinated patients)
