NAALADL2 antisense RNA 2 (NAALADL2-AS2) is a long non-coding RNA molecule located in both the cytoplasm and nucleus of cancer cells. It functions as a competing endogenous RNA (ceRNA), sequestering miR-34a and miR-125a, which leads to the upregulation of BCL-2, an anti-apoptotic protein, thus promoting cell survival and drug resistance in diffuse large B-cell lymphoma (DLBCL). Inhibition of NAALADL2-AS2 expression sensitizes DLBCL cells to chemotherapeutic drugs such as doxorubicin and rituximab by enhancing apoptosis. Its expression is also increased in metastatic castration-resistant prostate cancer and is associated with better response to enzalutamide, suggesting its relevance across different cancer types. Due to its regulatory role in apoptosis and drug resistance, NAALADL2-AS2 is being explored as a biomarker and a therapeutic target for overcoming chemotherapy resistance and improving patient outcomes in hematological malignancies and possibly solid tumors.
RNA interference and gene regulation: NAALADL2-AS2 inhibition increases sensitivity to chemotherapeutic drugs by promoting apoptosis through upregulating miR-34a and miR-125a, leading to downregulation of BCL-2, an anti-apoptotic protein.
