Overview

NAC-alpha domain-containing protein 1 (NACAD) is a cytoplasmic protein that is part of the nascent polypeptide-associated complex (NAC). It is predicted to bind nascent polypeptide chains as they emerge from the ribosome, preventing their inappropriate targeting to the endoplasmic reticulum by blocking interaction with the signal recognition particle (SRP). In this way, NACAD helps ensure that only appropriate secretory or membrane proteins are directed to the ER, thus regulating cellular protein trafficking. Disease associations are very limited, with only rare or possible links such as to cerebral cavernous malformations[1][3][4][7][10]. The protein is not a typical therapeutic target such as a receptor, enzyme, or transporter[1][3][4][7][10]. Its main molecular function is protein binding, specifically related to nascent (newly synthesized) polypeptide chains and their proper cellular targeting[1][7][10].
