NACHT and WD repeat domain containing 1 (NWD1) is a cytosolic protein classified within the STAND (Signal Transduction ATPases with Numerous Domains) protein family, characterized by the presence of a NACHT nucleotide-binding domain and multiple C-terminal WD40 repeats. Structurally and phylogenetically, NWD1 is related to both the NLR (NOD-like receptor) subfamily and Apaf1 (a key regulator of apoptosis). The protein is highly expressed in certain tissues, including the prostate and neural stem/progenitor cells. Functionally, NWD1 is a critical regulator of androgen receptor (AR) protein steady-state levels and activity, with its expression tightly correlated to prostate cancer progression and AR-mediated gene expression. It is upregulated by SRY-family transcription factors and essential for the maintenance of AR-dependent prostate cancer cell growth, in part through supporting PDEF, a transcription factor that co-regulates AR. Additional studies suggest roles in neuronal development, axonal growth, and possible contributions to cellular stress and apoptosis pathways, making it an emerging but atypical target involved in malignancy and neural biology
