Target intelligence / Profile preview
NAD+ biosynthesis pathway enzymes are a group of metabolic enzymes responsible for producing nicotinamide adenine dinucleotide (NAD+), an essential coenzyme in cellular redox processes, energy metabolism, DNA repair, and signal transduction. They include enzymes of multiple interconnected pathways: the de novo pathway (from tryptophan via indoleamine 2,3-dioxygenase [IDO] or tryptophan 2,3-dioxygenase [TDO], then quinolinate phosphoribosyltransferase [QPRT]), the Preiss–Handler pathway (from nicotinic acid via nicotinic acid phosphoribosyltransferase [NAPRT], NMNATs, and NAD synthase [NADSYN]), and the salvage pathway (from nicotinamide via nicotinamide phosphoribosyltransferase [NAMPT] and NMNATs). These enzymes are critical regulators of NAD+ homeostasis, cellular metabolism, and have implications in aging, inflammation, cancer growth, and neurodegenerative disease. They are of interest for therapeutic drug development both as targets for enzyme inhibition (e.g., NAMPT inhibitors in cancer) and for activation or substrate supplementation (anti-aging or neuroprotection). Note: For structured data, refer to individual enzymes within the pathway (e.g., "Nicotinamide phosphoribosyltransferase (NAMPT)", "Nicotinic acid phosphoribosyltransferase (NAPRT)", "Nicotinamide mononucleotide adenylyltransferase (NMNAT1/2/3)", "NAD synthase (NADSYN)" for higher scientific accuracy and specificity.
Inhibition of NAD+ biosynthetic enzymes (e.g., NAMPT inhibition) leads to NAD+ depletion, selectively targeting rapidly proliferating cells (cancer therapy) Supplementation with NAD+ precursors increases NAD+ pool and cellular energy, modulating aging, mitochondrial function, and recovery from injury
