Overview

Sirtuin 1 (SIRT1) mRNA is the transcript encoding the NAD-dependent protein deacetylase sirtuin-1, a key regulator of cellular metabolism, stress response, and longevity (UniProt: Q96EB6). The SIRT1 protein functions by removing acetyl groups from various histone and non-histone proteins, such as p53 and PGC-1alpha, thereby modulating gene expression and metabolic pathways (PubMed: 21383247). While most therapeutic efforts focus on activating the SIRT1 protein using small molecules, the SIRT1 mRNA itself serves as a target for knockdown strategies in conditions where SIRT1 is pathologically overexpressed, such as in certain malignancies (PubMed: 25605113). Targeting the mRNA using antisense oligonucleotides or siRNA allows for precise down-regulation of the protein product. This approach is being explored to sensitize cancer cells to chemotherapy or to study the fundamental roles of sirtuins in aging and metabolic diseases. However, therapeutic targeting of SIRT1 mRNA must account for the protein's complex, context-dependent roles in different tissues (PubMed: 11672531).
