Overview

NADH:quinone oxidoreductase 1 (NQO1), also known as DT-diaphorase, is a cytosolic flavoprotein enzyme that catalyzes the two-electron reduction of quinones to hydroquinones using NADH or NADPH. This prevents redox cycling and reduces oxidative stress. NQO1 also stabilizes proteins like p53. It is overexpressed in many cancers, making it a potential drug target, while deficiencies increase susceptibility to carcinogens.

Mechanism of action
