NADH:ubiquinone oxidoreductase complex I assembly factor 7 (NDUFAF7) is a mitochondrial protein required for the proper assembly and stabilization of complex I in the mitochondrial electron transport chain[1][3][5]. It functions as an arginine methyltransferase, symmetrically dimethylating Arg85 on the NDUFS2 subunit of complex I early in its assembly[1][3]. This methylation is essential for the formation and stability of early assembly intermediates of complex I, ensuring functionality of the mitochondrial respiratory chain and effective ATP production[1][3][5]. Mutations in NDUFAF7 impair complex I biogenesis and are associated with mitochondrial disorders such as pathologic myopia and fatal early embryonic development in model organisms[1][3][5]. NDUFAF7 is considered a critical enzyme in mitochondrial bioenergetics but is not currently a direct drug target.
